About BOVIGAM®
BOVIGAM® is an in vitro laboratory test for the diagnosis of Bovine Tuberculosis in cattle, sheep, goats, buffalo, bison and other bovidae. The test is unique on the market and yields highly reliable results. BOVIGAM® is used widely as an ancillary test to the tuberculin skin-test. Various studies describe the effective use of BOVIGAM® in combination with the skin test in Tuberculosis eradication programs.
BOVIGAM® is a blood-based assay of cell mediated immunity for the diagnosis of Bovine Tuberculosis infection in cattle. Animals infected with Mycobacterium bovis can be identified by measuring the cytokine interferon gamma (IFN-γ) against tuberculin, an antigen used to aid in the diagnosis of Tuberculosis infection. Tuberculin Purified Protein Derivatives (PPD) antigens are presented to lymphocytes in whole blood cultures and the production of IFN-γ from the stimulated T cells is detected using a monoclonal antibody-based sandwich enzyme immunoassay (EIA). Lymphocytes from uninfected cattle do not produce IFN-γ to tuberculin PPD antigens and hence IFN-γ detection correlates with infection.
Test procedure
The BOVIGAM® test involves two stages. In the first stage, blood samples are incubated overnight with antigen (e.g. tuberculin PPD) to stimulate the lymphocytes to produce IFN-γ. In the second stage, IFN-γ in the plasma supernatants of the each blood aliquot is determined using a sandwich EIA. IFN-γ present in the sample binds to antibodies to bovine IFN-γ bound to a solid support and is visualized with a second anti-bovine IFN-γ antibody labeled with an enzyme that generates a color signal. Color development is proportional to the amount of bound IFN-γ.
Performance
The Gamma-interferon assay has been approved by the Office International de Epizooties (OIE) as an ancillary test to confirm or negate the results of an intradermal skin test.
In 2002, the gamma interferon test was confirmed as a supplement for the skin test by the European Commission's Standing Committee on the Food Chain and Animal Health in Brussels.
