About the PrioCHECK[HOCH]®[/HOCH] Salmonella Ab bovine ELISAs
The PrioCHECK® Salmonella Ab bovine ELISAs are the first commercially available tests for monitoring and surveillance of Salmonella infections in cattle herds. The tests allow fast and versatile detection of anti-Salmonella antibodies in serum, plasma and milk samples. Both ELISAs follow an easy 4-step protocol. Results are available within 150 min, making the tests highly suitable for screening and monitoring programs for bovine salmonellosis.
Test procedure
The PrioCHECK® Salmonella Ab bovine is an indirect ELISAs for the detection of antibodies against the 1, 4, 5, 12 LPS O-antigens of the S. Dublin strain and 1, 9, 12 of the S. Typhimurium strain.
The PrioCHECK® Salmonella Ab bovine Dublin detects S. Dublin more specifically, but not exclusively: some cross-reactivity with S. Typhimurium can occur.
Both tests are suitable for large-scale screening of serum, plasma and (bulk) milk samples.
Test samples are incubated in plates coated with the purified Salmonella antigen at room temperature. Plates are then washed and an anti-bovine antibody labeled with an enzyme that generates a color signal is added. The signal is measured and if color develops the sample is positive for anti-Salmonella antibodies. Color intensity is directly related to the amount of anti-Salmonella antibody present in the test sample.
Performance
The PrioCHECK® Salmonella Ab bovine and the PrioCHECK® Salmonella Ab bovine Dublin were developed in cooperation with the Danish Veterinary Institute. The internal validation showed an exact correlation of the Prionics bovine Salmonella ELISAs with the in-house test of the Danish Veterinary Institute.
Internal and external validations showed an excellent performance of the PrioCHECK® Salmonella Ab bovine ELISAs. In the external validation conducted by the Swedish National Veterinary Institute on 611 negative and 203 positive bulk milk samples, the PrioCHECK® Salmonella Ab bovine Dublin showed a sensitivity and specificity of 99.4 % and 99.8 %, respectively.
