About the PrioCHECK® BTV DR
The PrioCHECK® BTV DR detects very low titers of anti-Bluetongue Virus (BTV) antibodies targeted to one of the major viral proteins of the Bluetongue Virus (BTV), VP7. This protein is highly conserved amongst BTV serotypes and a major immunogenic antigen ideal for BTV antibody detection. The PrioCHECK® BTV DR reveals the presence of anti-BTV antibodies as early as post infection day 6 in sheep and post infection day 7 in cattle. This versatile ELISA detects antibodies specific to the 24 serotypes known in the serum and plasma samples of cattle, sheep and goats. It can also identify immunized animals and can be therefore used to monitor the success of vaccination programs.
Test procedure
Prionics PrioCHECK® BTV DR is a diagnostic kit based on ELISA technology. The VP7 antigen of Bluetongue Virus (BTV) is detected twice, in the antibody capture, as well as in the detection step, where it is used as a conjugate – hence the name double recognition, DR.
The test follows a three step protocol consisting of sample incubation, conjugate incubation and detection. Serum or plasma samples are incubated in an ELISA plate coated with the VP7 antigen. Captured antibodies present in the sample are detected with the VP7 antigen conjugated to horse radish peroxidase (HRPO). Color development using tetramethyl-benzidine (TMB) substrate measured optically at a wavelength of 450 nm shows the presence of antibodies directed against BTV.

Performance
In a field trial testing 704 sera from herds certified as BTV-free, the PrioCHECK® BTV DR showed a specificity of 99.8%. Additionally, a set of 172 sera, previously characterized as positive, were analyzed. The ELISA recognized all 172 positive samples correctly, resulting in a sensitivity of 100%.
