About the PrioCHECK® HEV Ab porcine
The PrioCHECK® HEV Ab porcine is a reliable and fast diagnostic test for monitoring and surveillance of Hepatitis E virus in pig herds. The PrioCHECK® HEV Ab porcine is the only commercially available ELISA that is based on HEV genotype 1 and 3 antigens. The use of genotype 3 antigens – the HEV genotype that has actually been demonstrated to infect pigs – significantly enhances the specificity and sensitivity of the test. Sensitive and specific identification of infected animals allows reliable surveying and monitoring of the status of the virus in pig herds, leading to efficient implementation of appropriate measures in infected herds.
With the fast and robust PrioCHECK® HEV Ab porcine that follows a short four step protocol, 90 prepared samples can be analyzed within 150 minutes. The test thus allows cost-effective testing of large numbers of samples. The same sample taken for testing other pig diseases such as Salmonella, Toxoplasma or Trichinella infections can also be used in the PrioCHECK® HEV Ab porcine. The benefit: Tests can be easily combined and no additional sample has to be taken, allowing efficient addressing of the herd health status.
Test procedure
The PrioCHECK® HEV Ab porcine is a diagnostic kit based on ELISA technology. It follows a four step protocol consisting of sample preparation, sample incubation, conjugate incubation and detection.
Serum or meat juice samples are incubated in an ELISA plate coated with the recombinant HEV antigens 1 and 3. Anti-HEV antibodies present in the sample bind to the antigen. Plates are then washed and an anti-swine antibody labeled with horse radish peroxidase (HRP) is used to detect antibodies bound to the HEV antigen. Color development using tetramethyl-benzidine (TMB) substrate measured optically at a wavelength of 450 nm shows the presence of antibodies directed against HEV.
Performance
The PrioCHECK® HEV Ab porcine has shown a sensitivity of 91.0 % and a specificity of 94.0 % on 365 positive and 151 negative pig serum samples.* As reference method the recomLine HEV assay of Mikrogen GmbH was used, a commercial test for human diagnostics.
