PARACHEK® 2 is a solid phase, indirect enzyme immunoassay (EIA) to detect antibodies to M. paratuberculosis in animal sera and milk for the detection of paratuberculosis in cattle, sheep and goats. The test is a useful tool in the management and control of Paratuberculosis in conjunction with good farm management.
The technology on which this test is based has successfully overcome the problem of the cross-reactivity with antibodies to other mycobacteria unrelated to the disease, which is a significant serological problem of other tests. A rapid absorption step has been incorporated, early in the test procedure, to remove a range of cross-reacting antibodies produced in response to environmental mycobacteria. Following this rapid absorption phase, identification and detection of specific antibodies to M. paratuberculosis is possible.
Test procedure
The PARACHEK® 2 procedure for 96 determinations (including controls) takes approximately 2 hours and consists of four steps. In the first step, the samples are diluted and incubated in a diluent buffer containing M. phlei to remove cross reacting antibodies. In the second step, specific antibodies present in the diluted samples bind to M. paratuberculosis antigens coated to the wells of a microtiterplate. Unreacted proteins are removed by washing. A secondary antibody that is conjugated to an enzyme that generates a color signal is added in the third step. The presence of specific antibodies is then – in the fourth step – visualized by adding the enzyme substrate. The intensity of the color signal is proportional to the amount of antibodies in the sample.
Performance
Paratuberculosis occurs in multiple species and is a notifiable disease according to the OIE (Office International de Epizooties). PARACHEK® 2 is based on the existing PARACHEK® test, that has been used successfully in cattle, sheep and goats in many countries around the world.
